Morbidity and non-relapse mortality after allogeneic bone marrow transplantation in adult leukemia patients conditioned with busulfan plus cyclophosphamide: a retrospective comparison of oral versus intravenous busulfan.

چکیده
We retrospectively compared morbidity and non-relapse mortality (NRM) after allogeneic bone marrow transplantation (BMT) in 236 adults with leukemia and myelodysplastic syndrome conditioned with cyclophosphamide plus oral versus intravenous busulfan. Our findings demonstrate that conditioning therapy with intravenous busulfan resulted in lower morbidity and NRM than did oral busulfan.

Intravenous busulfan for autologous stem cell transplantation in adult patients with acute myeloid leukemia: a survey of 952 patients on behalf of the Acute Leukemia Working Party of the European Group for Blood and Marrow Transplantation.
Oral busulfan is the historical backbone of the busulfan+cyclophosphamide regimen for autologous stem cell transplantation. However intravenous busulfan has more predictable pharmacokinetics and less toxicity than oral busulfan; we, therefore, retrospectively analyzed data from 952 patients with acute myeloid leukemia who received intravenous busulfan for autologous stem cell transplantation. M...
متن کاملConditioning with Fludarabine-Busulfan versus Busulfan-Cyclophosphamide Is Associated with Lower aGVHD and Higher Survival but More Extensive and Long Standing Bone Marrow Damage
Acute graft-versus-host disease (aGVHD) is a major complication of allogeneic hematopoietic stem cell transplantation (allo-HSCT) and a major cause of nonrelapse mortality after allo-HSCT. A conditioning regimen plays a pivotal role in the development of aGVHD. To provide a platform for studying aGVHD and evaluating the impact of different conditioning regimens, we established a murine aGVHD mo...

Busulfan, a sulfonic acid ester, is an alkylating agent with potent toxicity for hematopoietic and sinusoidal endothelial cells. The drug was initially introduced in 1952, as a treatment modality for patients with chronic myeloid leukemia. It was only around 20 years later that this agent was first used as part of the conditioning regimen in bone marrow transplantation, substituting total body ...
متن کاملEffects of chimerism on graft-versus-host disease, disease recurrence, and survival after HLA-identical marrow transplantation in Iran.
BACKGROUND The coexistence of recipient's and donor's hematopoietic systems after allogeneic marrow transplantation is called mixed chimerism. OBJECTIVE The objective of this study was to evaluate the effects of MC on graft-versus-host disease (GVHD), disease recurrence, and survival after HLA identical marrow transplantation in a transplant center in Iran. METHODS The association of MC wit...
